Within the recently awarded STW Perspective Program: Understanding Processes using Operando Nanoscopy (UPON) we are looking for a 4 year PhD student at Maastricht University for project B4: “Bio-MEMS for structural biology using in-situ TEM.”

This PhD position is on a project in collaboration with Maastricht Instruments and NCHREM Delft, as part of the research program on the secretion machinery of mycobacterium species, and is focused on the purification certain proteins in combinations with EM sample preparation techniques such that subsequent data analysis is greatly facilitated. We will explore the use of MEMS nanochambers as tool for purification by applying electric fields for nanofluidic capillary electrophoresis and with subsequent TEM inspection. Initial tests will be done with fluorescent proteins with correlative light electron microscopy studies. We will also explore the use of caged heavy atom stains to facilitate detection of protein complexes at room temperature. The nanoreactors will be produced in a parallel project within UPON, an STW (www.stw.nl) and industry supported program consisting of 8 projects concentrating on the MEMS technique based development of nanoreactors and actual scientific and industrial applications (see the pdf). Modern TEMs are part of the infrastructure available.

Requirements:

  • master degree in biology or physics.
  • experience in transmission electron microscopy is a pre.
  • excellent writing and oral communication skills in English.
  • excellent collaboration skills (with other scientists and technicians in the larger program).
